Inhibition of the cytochrome P450 enzymes CYP3A4 and CYP2C19 is the primary mechanism of sildenafil-induced adverse events. Sildenafil is also a potent inhibitor of CYP3A4. In this study, we have identified two new CYP isoforms, isoforms X (also known as CYP1A2) and Xa (also known as CYP2C19), and we have determined the binding of the inhibitor and substrate of CYP isoforms X and Xa to the enzyme. The inhibition of CYP3A4 was determined using a substrate with a high affinity for the substrate and a low affinity for the substrate. The affinity of the inhibitor for the substrate was determined by comparing the Km values of the substrate with the Km values of the substrate. In vitro binding of the inhibitor to the enzyme was also determined in the presence of the substrate and the active-site. A single-site interaction study using the inhibitory properties of the CYP isoforms X and Xa was performed with a substrate and a P450 isoform inhibitor. The study showed that the inhibitors competitively inhibited the enzyme at the 10-mercaptoethanol level, while the inhibitor did not inhibit the enzyme at the 10-molecular weight level. These results were confirmed by the binding studies with the CYP isoforms. In vitro binding of the inhibitor to the enzyme was also determined in the presence of the inhibitor and in the presence of the P450 inhibitor. In addition, the affinity of the inhibitor for the substrate was determined by comparing the Km values of the inhibitor with the Km values of the substrate.
